Protein Sequencing & Analysis
The facility offers both N-terminal and C-terminal sequence analysis with interpretation for the novice or seasoned investigator. Microbore and capillary HPLCs, easily facilitate investigators desiring separation of micro-protein digests for N-terminal Sequencing or mass mapping by LC-ESI-MS. Purification at a wide variety of levels is available by HPLC, PAGE, and capillary electrophoresis for proteins, peptides, DNA, and small molecules. Molecular weight determinations of biomolecules are performed by direct infusion electrospray mass spectrometry(ESI-MS) and mass mapping by liquid chromatography electrospray mass spectrometry(LC-ESI-MS).
Complimenting the facility’s analytical abilities a BIACORE 2000 gives the researcher the ability to probe protein and DNA interactions with real-time monitoring without the use of labels. Data base searches and assistance with design of experimental protocols are available in the MSSSF. Sample preparation by facility staff is easily arranged by the investigator. The MSSSF maintains an online ordering system for oligonucleotides and its other services. Joseph Leykam can offer assistance with data base searching or oligonucleotide design.
The MSSSF is accessible to all investigators at Michigan State University and other institutions in the United States and abroad. The facility serves a diverse group of over 800 researchers throughout the world. We are committed to the training of faculty, staff, graduate students, and undergraduate students. Training on a variety of instrumentation is arranged by contacting Joseph Leykam.

History
The Macromolecular Structure, Sequencing and Synthesis Facility located in the Department of Biochemistry at Michigan State University was established by Dr. Jack Preiss in 1986 with the goal of providing timely cutting edge analysis of a variety of biomolecules. Equipped with a wide variety of instrumentation, the MSSSF analyses proteins, peptides, DNA, carbohydrates and small molecules. In addition, DNA synthesis is available with standard bases, modified bases, fluorescently labeled, or biotinylated oligonucleotides. Three synthesis levels and a variety of purification strategies are currently being offered. Oligonucleotide design can also be facilitated for the user through advanced analysis software prior to synthesis.
